چکیده
Abstract Lipids produced by oleaginous yeasts are considered as sustainable sources for the production of biofuels and oleochemicals. The red yeast Rhodosporidium toruloides can accumulate lipids to over 70% its dry cell mass. To facilitate lipid extraction, a recombinant β-1,3-glucomannanase, MAN5C, has been applied partially breakdown R. wall. In this study, NP11 was engineered secretory expression MAN5C simplify extraction process. Specifically, cassette contained codon-optimized gene integrated into genome Agrobacterium -mediated transformation. strain NP11-MAN5C found with proper secretion active yet no notable compromise in terms growth production. When cultures were extracted ethyl acetate without any pretreatment, 20% total recovered, 4.3-fold higher than that parental NP11. cells heat-treated followed presence culture broth supernatants, up 93% confirming beneficial effects situ. This study provides new strategy engineer more viable down-stream processes. Graphical

Functional genomics of lipid metabolism in the oleaginous yeast Rhodosporidium toruloides.
The basidiomycete yeast Rhodosporidium toruloides (a.k.a. Rhodotorula toruloides) accumulates high concentrations of lipids and carotenoids from diverse carbon sources. It has great potential as a model for the cellular biology of lipid droplets and for sustainable chemical production. We developed a method for high-throughput genetics (RB-TDNAseq), using sequence-barcoded Agrobacterium tumefac...
متن کاملDynamics of the lipid droplet proteome of the Oleaginous yeast rhodosporidium toruloides.
Lipid droplets (LDs) are ubiquitous organelles that serve as a neutral lipid reservoir and a hub for lipid metabolism. Manipulating LD formation, evolution, and mobilization in oleaginous species may lead to the production of fatty acid-derived biofuels and chemicals. However, key factors regulating LD dynamics remain poorly characterized. Here we purified the LDs and identified LD-associated p...

We report the de novo assembled 20.05-Mb draft genome of the red yeast Rhodosporidium toruloides MTCC 457, predicted to encode 5,993 proteins, 4 rRNAs, and 125 tRNAs. Proteins known to be unique to oleaginous fungi are present among the predicted proteins. The genome sequence will be valuable for molecular genetic analysis and manipulation of lipid accumulation in this yeast and for developing ...

Microbial lipid production by the oleaginous yeast Rhorosporidium toruloides Y4 was studied using glucose as carbon source, in order to realize igh-density cell culture. Batch cultures demonstrated that there was little inhibitory effect with a substrate concentration of up to 150 g l−1. Flask ed-batch cultures were run for 25 days and reached a dry biomass and cellular lipid content of 151.5 g...
